If you manually adjust the Price at the main Product page, it will overwrite the discount rules and replaces the manually adjusted price. Please see the following screenshot for the example:

If the price is manually adjusted at the Price field:

It will display the manually adjusted price at the store:

The same also applies if Compare to Price field is manually adjusted.
